If you need a proxy to reach the Internet make sure that it is configured. From the screen shot i can see "Automatically detect settings". You can try to configure with no proxy or manually put your proxy to make sure.

Also you can look at the neoload.log file accessible trough the menu "Help-->Open logs folder" for more details about the connection error.
